Whole Genome
Samples are run with the Genomic Tape on the Agilent Tapestation 4200 and their concentrations quantified by fluorometery. One microgram genomic DNA is sonicated using Covaris E220 ultrasonicator targeting 400bp. Libraries are prepared with the KAPA Hyper Prep Kit. Samples are end repaired, 3' ends adenylated and barcoded with multiplex adapters. Samples are size selected prior to PCR to remove small fragments. Two cycles of PCR are used to amplify the libraries, which are then purified with AmpureXP beads and validated on the Agilent Tapestation 4200. Before being normalized and pooled, samples are quantified by Qubit then run on the appropriate Illumina instrument.
