QIAGEN AUTOPURE LS® – Large Sample Nucleic Acid Purification Instrument
The QIAGEN AUTOPURE LS® Automated Nucleic Acid Purification Instrument is a laboratory workstation designed to provide automated nucleic acid purification from large sample volumes for high-throughput research applications. It can process up to 96 samples in eight hours while providing complete sample tracking. We routinely obtain ~300µg of archive-quality genomic DNA from the buffy coat of a 10mL of whole blood.
Among the many features of the QIAGEN AUTOPURE LS® are consistent sample identification and tracking, a robotically accessible centrifuge, automated mixing, weighing, pouring and tube-to-tube transfer. In addition, specially designed processing tube are easily capped, uncapped, and transported throughout the instrument using a combination of robotic arms and transfer racks.
Genomic DNA Extraction
Blood samples in purple top EDTA tubes should be kept at 4oC. Blood from multiple tubes are combined to obtain a buffy coat from which genomic DNA is isolated. The DNA pellet is rehydrated with 500µL of DNA hydration solution. Only one tube of DNA will be returned for each order of genomic DNA.
- For samples of 10–20 mL of whole blood, the buffy coat(s) will be combined and processed by the automated AUTOPURE LS DNA purification instrument (www.qiagen.com). The DNA pellet will be rehydrated in 500 µL of DNA hydration solution.
- For samples less than 10 mL of whole blood, the entire sample will be processed by hand. The DNA pellet will be rehydrated in 250 µL of DNA hydration solution.
Buccal swabs should be stored at room temperature. The DNA pellet will be rehydrated in 50 µL of DNA hydration solution.
Oragene Spit Kit
Oragene Spit Kit samples should be stored at room temperature; they are stable for up to 2 years. The DNA pellet will be rehydrated in 400 µL of DNA hydration solution. For more information on Oragene products, please visit www.dnagenotek.com or contact firstname.lastname@example.org.
Only one (1) aliquot of DNA is processed and stored per sample. All DNA will be stored at 4oC (short-term) or -20oC (long-term).
